Multi-column chromatography is a bioprocessing technique where multiple columns are loaded in series and then cycled for elution, regeneration and equilibration. This allows fuller utilisation of the potential resin binding capacity, rapid cycling of much smaller columns and with careful scheduling a continuous elution stream suitable for continuous or connected bioprocessing applications. Whilst very effective at increasing operational efficiency, especially with Protein A primary capture of monoclonal antibodies, this technique does present challenges regarding system design, automation and control due to the increased complexity of the single-use flow path.
